The pathologic changes during graft-versus-host reaction were studied in normal, adrenalectomized and/or thymectomized (C3H × C57B1)F1 hybrid mice injected with spleen cells from C3H strain donors. Our findings suggest that lymphoid tissue damage during graft-versus-host reaction can be attributed to primary pathology of the thymus-dependent ‘T’ lymphoid tissue. The primary target of the immunologic graft-versus-host reaction appears to be the ‘T’ lymphoid component. The secondary pathology seems to be mediated by at least two mechanisms, adrenal gland hypersecretion and an adjacent tissue effect that may be dissected by manipulation of the host.
