Mouse skin was treated electrophoretically at 40 mA for 5 h. Two lipid-protein fractions were extracted from the supporting electrophoresis sheet. Fraction I, comprising 6 different protein bands in disc electrophoresis, was shown to have a specific antigenic property against homologous skin grafts. The MST decreased significantly from 11.2 days for the controls to 8.3 days when 0.1 mg of fraction I were injected subcutaneously 3 times before grafting. Fraction II had no effect. The antigenic substances are thought to originate from epidermal cells. An electrophoretic pattern comparable to that of fraction I was obtained by milling the skin.
