Cytochemical studies of various enzymes were carried out on megakaryocytes obtained from patients with disorders of megakaryocytes. In patients with idiopathic thrombocytopenic purpura megakaryocytes have uniformly low levels of the enzymes studied, as estimated by formazan deposition. In contrast, megakaryocytes obtained from patients with agnogenic myeloid metaplasia and especially essential thrombocythemia showed elevated enzymatic activities, particularly those of glycolytic enzymes such as lactic dehydrogenase. These findings presumably reflect the neoplastic nature of these myeloproliferative megakaryocytes.
